Doublecortin Is a Highly Valuable Endogenous Marker of Adult Neurogenesis in Canaries

چکیده
Vellema et al. [2014] report that DCXexpressing cells, based both on immunohistochemistry for the protein and in situ hybridization for the corresponding mRNA, are found throughout the canary brain, with the most pronounced expression in the telencephalon, predominantly in the mesopallium and caudal nidopallium. Based on other methods, this does indeed correspond to brain areas where an active incorporation of new neurons is observed in adult birds. The authors indicate that scattered labeled neurons were also found ‘occasionally’ in other regions outside the telencephalon (e.g. in the ventral tegmental area, the substantia nigra and the locus ceruleus). Endogenous versus exogenous markers of adult neurogenesis in canaries and other birds: advantages and disadvantages.
Although the existence of newborn neurons had originally been suggested, but not broadly accepted, based on studies in adult rodent brains, the presence of an active neurogenesis process in adult homoeothermic vertebrates was first firmly established in songbirds. Adult neurogenesis was initially studied with the tritiated thymidine technique, later replaced by the injection and detection of th...
متن کاملStudies of HVC Plasticity in Adult Canaries Reveal Social Effects and Sex Differences as Well as Limitations of Multiple Markers Available to Assess Adult Neurogenesis
In songbirds, neurogenesis in the song control nucleus HVC is sensitive to the hormonal and social environment but the dynamics of this process is difficult to assess with a single exogenous marker of new neurons. We simultaneously used three independent markers to investigate HVC neurogenesis in male and female canaries. Males were castrated, implanted with testosterone and housed either alone...
متن کاملWhat Makes a Marker a Good Marker?
To validate if DCX truly predicts neuron recruitment throughout the canary brain, it should correlate to a high degree with established methods for studying neuron recruitment, such as labeling cells with the mitotic marker 5-bromo-2-deoxyuridine (BrdU). This is not the case, however, as we observed clear regional differences in the relationship between the two labeling methods in neurogenic an...
متن کاملRepeated PTZ Treatment at 25-Day Intervals Leads to a Highly Efficient Accumulation of Doublecortin in the Dorsal Hippocampus of Rats
BACKGROUND Neurogenesis persists throughout life in the adult mammalian brain. Because neurogenesis can only be assessed in postmortem tissue, its functional significance remains undetermined, and identifying an in vivo correlate of neurogenesis has become an important goal. By studying pentylenetetrazole-induced brain stimulation in a rat model of kindling we accidentally discovered that 25±1 ...
متن کاملEvidence for reduced neurogenesis in the aging human hippocampus despite stable stem cell markers
Reduced neurogenesis in the aging mammalian hippocampus has been linked to cognitive deficits and increased risk of dementia. We utilized postmortem human hippocampal tissue from 26 subjects aged 18-88 years to investigate changes in expression of six genes representing different stages of neurogenesis across the healthy adult lifespan. Progressive and significant decreases in mRNA levels of th...
